MDAI (5,6-methylenedioxy-2-aminoindane; 6,7-dihydro-5H-cyclopenta[f][1,3]benzodioxol-6-amine;’mindy’) is a psychoactive chemical that is predominantly offered as a ‘legal high’ via the Internet and in ‘head’ shops. MDAI was synthesized and utilized as a research chemical in the 1990s, and it exhibits structural and behavioral similarities to MDMA (3,4-methylenedioxy-N-methylamphetamine). MDAI use for recreational purposes appears to have begun in Europe around 2007, with a substantial increase in the UK and other countries after 2009.
Although there were few presentations to emergency rooms by patients complaining of unwanted physical and behavioral consequences after taking MDAI, calls to National Poisons Information Services began in 2010. Online user forums have only sporadically reported recreational use of this medication. The pharmacological, physiological, psychopharmacological, toxicological, and epidemiological features of this medication have minimal scientific support.
MDAI has been proven in study models to strongly enhance the release of the neurotransmitter serotonin, and hence can be defined as a’selective serotonin releasing agent’ (SSRA). Serotonin (also known as 5-HT) is mostly present in the intestines and central nervous system, where it regulates a variety of functions including bowel movement, sleep, mood, appetite, memory, and learning.
MDAI promotes serotonin release to a similar level as MDA, but less so than 3, 4-methylenedioxy-N-methylamphetamine (MDMA).
MDAI has been demonstrated to increase feelings of empathy, affection, and emotional connectedness with others in humans. Entactogens (or empathogens) are substances that produce these effects, and they include MDMA and related ‘MDxx’ medicines in addition to MDAI.
MDAI has also been discovered to impede serotonin reuptake (migration back into cells), as well as (to a lesser extent) the neurotransmitters dopamine (associated in regulating reward-seeking behavior) and norepinephrine (or noradrenaline – a key stress hormone).
